Washington's magic number down to 1 after Nationals knock off St. Louis Cardinals

ST. LOUIS ? Michael Morse circled the bases for a grand slam after taking an imaginary swing, and the Washington Nationals cut their magic number for winning the NL East to one Saturday night, beating St. Louis 6-4 on Kurt Suzuki's two-run double in the 10th inning.

The Nationals have put on hold a champagne celebration until they win the division, and were forced to wait at least another day when second-place Atlanta beat the Mets 2-0.

Washington had already assured the nation's capital of its first postseason appearance since 1933. The Nationals can win their first division title on Sunday when a St. Louis native, lefty Ross Detwiler, starts against Cardinals 17-game winner Lance Lynn.

St. Louis' magic number for clinching the second NL wild-card spot remained at three.

The Nationals opened the game with a blast, and a bit of comedy.

Bryce Harper singled, Ryan Zimmerman doubled and Adam LaRoche drew a full-count walk to load the bases in the first inning.

Morse hit a line drive on the first pitch that cleared the right-field wall and landed in a grassy section beneath a video board. First base umpire Chris Guccione didn't see it, however, and play continued. Right fielder Carlos Beltran made a relay to second baseman Skip Schumaker, who ran down Morse as he retreated toward first.

The umpires looked at video replays and confirmed it was indeed a home run. The umps ordered all runners to retrace their steps and put Morse back at the plate, where he mimicked his swing minus a bat and then made his trot.

Morse has four career slams and two this season. He is 6 for 11 with 14 RBIs with the bases loaded this year.

Pete Kozma had three hits and two RBIs for St. Louis. He scored on Jon Jay's sacrifice fly off Drew Storen (3-1) in the ninth to tie it.

The Nationals got a pair of walks and a passed ball to set up the go-ahead hit, a double off the wall in left-center by Suzuki, who's batting .385 with runners in scoring position.

LaRoche walked for second time to open the 10th against rookie Sam Freeman (0-2) and also had a pair of hits.

Craig Stammen allowed a hit but also got a double-play ball in the 10th for his first save in two chances.

Just like a night earlier when the Nationals' Edwin Jackson trailed 5-1 after an inning, Cardinals starter Kyle Lohse got into deep trouble early.

Nationals starter Jordan Zimmermann entered 0-2 with a 10.71 ERA in four starts against the Cardinals, allowing at least five runs each time. He retired eight of nine to open the game and before the seventh had allowed only one runner into scoring position.

The Cardinals got three straight one-out hits to chase Zimmermann in the seventh, singles by Beltran and Skip Schumaker ahead of a two-run double by Kozma, the eighth-place hitter. Jay added an RBI single off Sean Burnett to cut the deficit to one.

NOTES: The blown save was Storen's first in four chances. ... Cardinals 3B David Freese missed his fifth straight start with a sprained right ankle but could play Sunday. Matt Carpenter subbed for him again Saturday. Freese drew a five-pitch walk as a pinch hitter in the seventh. ... Beltran, who entered in a 3 for 21 slump, batted sixth for the second straight night and fourth time overall this season. He's been the primary cleanup man and has 92 RBIs, but just 27 since the All-Star break.

Promising new data from trials aimed at delaying resistance to BRAF inhibitors

Vienna, Austria, 29 September 2012 - Promising new data on drug combinations to treat metastatic melanoma are presented at the ESMO 2012 Congress of the European Society for Medical Oncology in Vienna.

The phase I and II trials focus on combining drugs to slow the development of resistance to drugs that inhibit BRAF, a gene that is mutated in about half of melanomas. Earlier trials with drugs that target BRAF generated excitement for their ability to quickly shrink melanoma tumors in suitable patients. But for many patients the benefits proved short-lived, as the cancer cells develop resistance to the drugs.

"These studies exemplify an important landmark of some tumors, which has emerged from recent laboratory research: the presence of specific mutations, such as the BRAF mutation in metastatic melanoma which exposes an Achilles' heel--MEK in this case," said Prof Yossef Yarden from the Weizmann Institute of Science, Israel. "In-depth understanding of cancers and their mutations is expected to reveal more of these deadly weaknesses in cancer, which we can exploit using new drugs and drug combinations."

Phase II of the BRAF inhibitor dabrafenib alone vs combination with MEK1/2 inhibitor trametinib

Dr Georgina Long from Westmead Hospital and the Melanoma Institute Australia and colleagues report that combining the new drugs dabrafenib and trametinib provided a clinically meaningful improvement in progression-free survival, response rate and duration of response in 162 patients with melanoma that had BRAF V600 mutations.

Patients in the study received either dabrafenib 150mg twice daily; twice-daily dabrafenib plus once-daily 1mg trametinib; or twice daily dabrafenib plus once-daily 2mg trametinib. The combination prolonged progresion free survival over single-drug therapy from 5.8 months to 9.4 months, which represents a 60% improvement. Among patients who received both drugs at the higher dose, 41% had not progressed 12 months after treatment began, compared to 9% in the monotherapy arm of the study.

"The combination therapy of the BRAF inhibitor dabrafenib and the MEK inhibitor trametinib prolongs the progression-free survival in patients with V600 BRAF mutation-positive metastatic melanoma compared with dabrafenic monotherapy," Dr Long said. "Importantly, the combination also decreases the rate of the cutaneous toxicities compared with dabrafenib monotherapy, particularly the oncogenic cutaneous toxicity of squamous cell carcinoma."

Phase IB study of vemurafenib in combination with the MEK inhibitor, GDC-0973

A Phase I study in 44 patients shows that the combination of the MEK inhibitor GDC-0973 and vemurafenib can be delivered safely, Dr Rene Gonzalez of the University of Colorado Cancer Center, Denver, and colleagues report.

"BRAF inhibition has resulted in high response rates and improved survival in patients with BRAF mutated melanoma," Dr Gonzalez said. "One of several mechanisms of resistance has been reactivation of the MAPK pathway. Preclinical models show that combined inhibition of BRAF and MEK can delay the acqusition of resistance compared to BRAF inhibitor monotherapy. Inhibition of the pathway downstream from BRAF with the MEK inhibitor GDC-0973 could theoretically overcome or delay this resistance mechanism and improve outcomes."

The study was not designed to evalate efficacy. "While early data in a small number of patients did show tumor reduction, it would be premature to comment on efficacy based on these preliminary results and further research is warranted," Dr Gonzalez said.

Nuclear site ends security contract following nun's break-in

WASHINGTON (Reuters) - The government's "Fort Knox" of weapons-grade uranium storage has ended a contract with a unit of an international security firm two months after an 82-year-old nun and other nuclear activists broke into the site.

The managing contractor at the Oak Ridge, Tennessee, site, B&W Y-12, a unit of Babcock & Wilcox Co, said late on Friday it will terminate the contract with WSI Oak Ridge on October 1. WSI is owned by security firm G4S, which was at the center of a dispute over security at this year's London Olympic Games.

The move came after the National Nuclear Security Administration (NNSA), an Energy Department agency, sent a letter on Friday to B&W Y-12 President Charles Spencer saying it had "grave concerns" about his company and WSI providing security at Y-12, the nation's only site for storing and processing weapons-grade uranium.

The letter recommended that B&W terminate the subcontract with WSI and work with it to take over security operations after the July 28 break-in.

The nun, Megan Rice, and two others cut perimeter fences to reach the outer wall of a building where enriched uranium was stored. The site was shut temporarily after the breach.

An investigation by the Energy Department's inspector general last month found a security camera had been broken for about six months and was part of a backlog of repairs needed for security at the facility.

The NNSA repeated on Saturday that Energy Secretary Steven Chu has said the incident was an important "wake-up call" for the entire nuclear complex.

"The security of our nation's nuclear material is the Department's most important responsibility, and we have no tolerance for federal or contractor personnel who cannot or will not do their jobs," said NNSA spokesman Joshua McConaha.

After the incident the NNSA's top security official and two other federal officials were reassigned. In addition, top officials at WSI were removed and officers associated with the break-in were fired, demoted, or suspended without pay.

WSI's parent company, G4S, found itself the focus of a political and media storm this summer in Britain over outsourcing of security after it failed to provide enough guards for the Olympics.

WSI did not immediately answer a request for comment about the ending of the contract.

It seems few other jobs will be lost over the incident that brought new questions about the government's outsourcing of sensitive security operations.

B&W said in a statement it will offer employment to all Y-12 security police officers and active union workers with WSI Oak Ridge.

The NNSA and Department of Energy are engaged in reviews of security operations from the contractor, to the federal management, to the security model, McConaha said.

The final review will begin after Chu asks outside observers to analyze current protection of nuclear materials and explore more options for protecting the sites.

Chu received a classified review of the Y-12 incident earlier in the week by the department's health, safety, and security office.

You may note that Arkansas was previously thoroughly dismantled by the likes of Alabama (52-0), Rutgers (35-26) and Louisiana-Monroe (an OT victory in a game Arkansas was supposed to win by 30). In that, you would be right. Just how can a team that has shown only bare flashes of being mantled be yet again dismantled? Oh, when it gives up 58 points to the Aggies, that's how. Also when it scores only 10 points on 515 yards of total offense. Arkansas is no longer a college football team; it's an inflatable bouncy castle that kids jump on until they get tired and a little sweaty and then go home laughing. At least the Hogs don't apply postage stamps when they send faxes, as Aggies have been known to do.

But back to Manziel and his offensive outburst. Almost a third of a mile, this fellow accounted for! He threw for 453 yards and three touchdowns. He ran for 104 yards and a touchdown on just 14 carries, including a 52-yard scamper that made the Arkansas secondary look bewildered, slow and timid. His output bettered Archie Manning's 1969 game against Alabama and Rohan Davey's 2001 game against Alabama - both 540-yard days - as the best in conference history. It was altogether mindblowing, like when you put an Aggie in a round room and tell him to stand in the corner.

Of course, Manning as an Ole Miss quarterback and even Davey, as an LSU QB, can stake a claim to a longer SEC heritage. To see an SEC record to be set by an A&M player - and against Arkansas, no less! It almost seems wrong the watch two SWC rivals combine to unseat a Manning from SEC record books. Almost as wrong as hanging your A&M diploma on the rearview to justify parking in handicapped spots.
